Letter to the Editor: Hypothalamic Kisspeptin in Female Rat Models of PCOS

A recent article by Osuka et al. (1) reports the use of two female rat models of polycystic ovary syndrome (PCOS) to explore the effects of prenatal or postnatal androgenization on the estrous cycle, hormonal profiles, and expression of kisspeptin in the hypothalamus. Their interesting data suggest that prenatal, but not postnatal, treatment with dihydrotestosterone (DHT) significantly elevated the number of kisspeptin-positive cell bodies in the rostral and middle arcuate nucleus. However, there was no effect of prenatal androgenization on kisspeptin immunoreactivity or on kiss1 messenger RNA (mRNA) in the anteroventral periventricular nucleus. Of note, an earlier report (2) used an identical rat model of PCOS (3) to investigate the effects of postnatal (prepubertal) treatment with DHT on hypothalamic kiss1 mRNA and on kisspeptin immunoreactivity. This study demonstrated a profound DHT-induced reduction of kisspeptin immunoreactivity in the arcuate nucleus and anteroventral periventricular nucleus at postnatal day 47, and this depletion persisted at postnatal day 81. This is in contrast to the finding of Osuka et al. (1) that postnatal DHT treatment had no effect on arcuate nucleus kisspeptin immunoreactivity. The conflicting data obtained from these two independent investigations, using the same postnatal DHT treatment model, merit a comparison of the studies. There are some differences in the experimental approaches (e.g., the use of Wistar vs Sprague-Dawley rats, and in the antibodies used for immunohistochemistry), by which alternative data may accrue as a result. However, it is noteworthy that the postnatal DHT-induced effects on body weight, vaginal opening, ovarian weight, and the reduction in numbers of corpora lutea were almost identical in the two reports, suggesting that DHT treatment in both rat strains was similarly effective and comparable to the data originally published by Mannerås et al. (3). One of the characteristics of human PCOS is an enhanced pulsatile secretion of luteinizing hormone (LH) (4). Given the well-described stimulatory influence of kisspeptin on gonadotropin-releasing hormone and LH secretion in rodents (5), our findings of a significant DHT-induced decrease in hypothalamic kisspeptin immunoreactivity and kiss1 mRNA (2) suggest that a corresponding increase in LH secretion in this model is unlikely. The possibility remains, therefore, that female rats treated postnatally with DHT may not represent an adequate model of human PCOS. Indeed, despite the availability of several PCOS models (6), it remains important to acknowledge their limitations, especially in rodents.
