Soluble fiber beta-glucan from oats at higher concentrations promotes hydrogen peroxide-induced cytotoxicity in small intestine epithelial cells

In order to determine whether b -glucan from oats can shield intestine epithelial cells from oxidative cell injury, rat small intestine epithelial cells (IEC-6) were cultured with or without medium viscosity oat b -glucan at concentrations of 0.1, 0.5, 2 and 4 mg/ml and treated with serial diluted H2O2. For the enzyme activity and protein expression assays, cells were cultured with 2 or 4mg/ml b -glucan and applied with 8uM H2O2. There is no difference in cell survival in response to the H2O2 treatment when cells were cultured with 0.1 or 0.5 mg/ml of b -glucan as compared with those cultured without b -glucan. However, when 2 or 4 mg/ml of b -glucan is applied to the cells, low concentrations of H2O2 (3.9 to 15.5 μM), which are not toxic to the cells growing without added b -glucan, become poisonous to the cells. Superoxide dismutase activity was increased by a combined application of H2O2 and b -glucan or b -glucan alone. The expression of TNF-a was decreased in cells co-cultured with either H2O2 or b -glucan. A combination of H2O2 and b -glucan restored the expression of this protein to the level in the non-treated control cells. The activity of caspase-3 was increased by H2O2 only when the higher concentration of b -glucan is present. In summary, low levels of b -glucan do not appear to protect small intestine epithelial cells against H2O2 induced oxidative injury. On the contrary, the presence of higher concentrations of this soluble fiber produces an oxidative-stress like challenge and promotes hydrogen peroxide-induced cytotoxicity to those cells.
