The infection caused by the coronavirus type 2 (SARS-COV-2), started in late 2019 in Wuhan, China and in a few months spread to several countries, becoming a pandemic.In Brazil, the first case occurred at the end of February 2020, and in nine months it infected more than 5.5 million people and totaled more than 160 thousand deaths.The virus infection can progress asymptomatically to severe symptoms, such as severe acute respiratory syndrome.The laboratory features described in patients with Covid-19 are the presence of neutrophilia and lymphopenia, lymphocyte exhaustion and increased levels of pro-inflammatory cytokines.The damage caused by neutrophilia has been investigated in Covid-19, however, observations on how to attenuate neutrophil-mediated oxidative stress are lacking.In this study, the proposal was to evaluate neutrophils and their functional capacity in patients with moderate, severe and critical Covid-19, hospitalized at HC-FMUSP, and the antioxidant capacity of Resveratrol.190 patients with Covid-19 were included, 108 with moderate symptoms, 21 severe and 61 critical, during the period from May 2020 to October 2021.Regarding neutrophils, in addition to neutrophilia, an increase in neutrophils with characteristics immature cells characterized by the absence of CD10 marker expression.This immature characteristic was verified in prevalence in patients with severe and critical symptoms.In the functional analysis, a higher oxidative capacity of neutrophils in moderate patients compared to healthy individuals was verified, but reduced according to the severity of the disease.In the TSNE evaluation, we identified a cluster in the population of neutrophils only in patients with Covid-19 and absent in individuals in the control group.Resveratrol-mediated antioxidant action was evidenced by the decrease in the oxidative response of neutrophils, the formation of Neutrophil Extracellular Traps (NETs) and the levels of cell-free DNA in the culture with neutrophils.Taken together, the data show that despite the neutrophilia and the oxidative potential in SARS-Cov-2 infection, there is a reduction in the oxidative index, due to the probable presence of immature neutrophils.Resveratrol's antioxidant action suggests that it is a potential candidate to attenuate the harmful effects of neutrophils in respiratory viral infections such as Covid-19 infection.
