Abstract 283: Expression of the Pan-Neurotrophic Receptor p75 <sup>NTR</sup> in Human Platelets

Background: The brain-derived neurotrophic factor (BDNF) is a protein from the neurotrophin family. This peptide was first identified in the central nervous system (CNS), where it is implicated in learning and memory by promoting neuron survival through the tropomyosin receptor kinase B (TrkB) as well as by acting on the 75 kDa pan-neurotrophic receptor (p75 NTR ), which can mediate both neuron survival and apoptosis. Despite being discovered in the brain, BDNF is more abundant in blood, where its concentrations can reach 100 to 1000 times the ones found in the brain. In blood circulation, BDNF is contained within platelets, making this cell type the largest peripheral reserve of this peptide. It is known that BDNF is secreted from platelets upon activation and preliminary results obtained in our lab show that physiological amounts of BDNF induce both activation and complete platelet aggregation. However, the underlying mechanism and the protein acting as a receptor for BDNF at the platelet membrane are still unknown to this day. Hypothesis: We hypothesised that the receptor activating the whole pathway mediating aggregation was the p75 NTR , known to be bound by the BDNF protein in the CNS. Methods and results: We first determined the presence of the receptor on platelets by western blotting. Similarly to human cortex cell lysates, platelet lysates displayed a clear band at the expected molecular weight of 75 kDa for p75 NTR . We then characterized its location using flow cytometry and confocal microscopy, which revealed that the receptor is present both at the membrane and within platelets. Furthermore, by co-immunoprecipitation, we demonstrated that the p75 NTR receptor is bound by BDNF in platelets of healthy volunteers, thus showing an interaction between these two peptides at the platelet level. Conclusion: We demonstrated for the first time that platelets express the pan-neurotrophic receptor, p75 NTR . The expression pattern of p75 NTR suggests a role either in the mediation of BDNF signalling leading to platelet aggregation or in the internalisation of BDNF by platelets. This warrants further investigation into the physiological relevance of the BDNF-p75 NTR loop in platelets and vascular homeostasis.
