Symmetry applied to nuclear microanatomy: a review of gene function and cell differentiation

The purpose of this paper is to review current \nknowledge and understandings of gene control and cell \ndifferentiation, based upon an appreciation of a possible \nrole that nuclear microanatomy and considerations of \nsteric symmetry might play. \nMetaphase sister chromatids have identical base \ncodes but show a mirror image symmetry of higher order \ncoiling. Chromosomes in the interphase nucleus have \nspatially well defined domains and are anatomically \ndistinct and ordered. Chromosomes are known to have \ninteractions i.e. sex chromosome inactivation, PEV etc \nAn hypothesis of gene activation is made based on \nsteric interactions among chromosomes and between \nchromosomes and activating and repressor proteins. \nThese interactions may be influenced by the handedness \nof higher order chromatid coiling, since homologues \nshow mirror-image symmetrical coiling in metaphase, \nwhich might be retained to a certain degree in \ninterphase. This may result in a binary switching of \ngenes. \nAll possible combinations of chromatids in the \ninterphase nucleus, would be enabled by a differential \nsegregation of homologous chromatids at mitosis. To \nconserve patterns of interchromatid interactions, there \nmust be a programmed segregation of chromatids \ntowards one of the two spindle pole attachments. This \norientation might be effected by preferential attachment of microtubules to kinetochore attachment sites, by steric \nhindrance of the kinetochore by condensed chromatin \nwhich initially allows only unidirectional tubule \nattachment, or possibly by a tethering of interacting \nchromatids which would migrate en masse. \nAn attempt to apply this hypothesis to some \nillustrative pathological conditions is made.
