The Role of the Cuticular Hydrocarbon Compounds, Produced by the Oenocytes, in Desiccation Resistance in Drosophila melanogaster

Terrestrial insects are susceptible to desiccation, due to their relatively small size, restricting their ability to maintain water balance. This limitation arises from the relatively large surface area to small volume. The physiological and behavior mechanisms employed to maintain water balance and protect against desiccation are central components to insect ecology. To withstand desiccation, insects have evolved diverse strategies, such as increasing water content, tolerating dehydration, and reducing water loss rate. Waxy hydrocarbon compounds on the outer surface of the insect cuticle form an impermeable barrier that is widely claimed to protect against desiccation. Despite extensive exploration of cuticular hydrocarbon compounds (CHCs) though various biochemical and genetic techniques across many insect species, the protective effects of these compounds have not been directly demonstrated. To address this gap, we establish a model system of transgenic strain of Drosophila melanogaster, specifically targeting the elimination of cuticular hydrocarbons by ablating the oenocytes. This approach allows us to assess the efficacy of CHCs in protecting against desiccation in the vinegar fly, Drosophila melanogaster. Using this system, we demonstrate that the oenocytes and CHCs produced by these cells are critically important for desiccation resistance, as measured by reduced survival under desiccative conditions. The underlying physiological mechanisms contributing to this reduced survival is increased evaporative water loss rate, irrespective of body water content and dehydration tolerance. Artificially coating these oenocyte-less (oe-) flies with fly-derived CHC extract or with mixtures of synthetic substitutes demonstrates that both straight-chain alkane and methyl-branched alkane compounds contribute to desiccation resistance. Notably, a combination of different compound classes, each possessing distinct physical and chemical properties, is essential for achieving maximal protection. Furthermore, our findings reveal that hydrocarbon compounds transferred to flies from an opposite-sex partner during copulation provide a short-term reduction in water loss rate and increase in desiccation survival. Mated females receive an additional longer-term benefit in desiccation resistance, lasting days, that is independent of CHC transfer. Together, CHCs and life history events, including changes in sexual status and environmental temperature, play important roles in influencing physiological changes that increase protection to desiccation, serving to offset the survival costs associated with sexual reproduction.
