Ageing alters hypertension‐ and sex‐related influences on endothelial vasomotor function in rat thoracic aorta

Abstract
Sex-specific vascular adaptations to hypertension and ageing were examined in young (16wk) and adult (30wk), male (M) and female (F), normotensive Wistar-Kyoto (WKY) and spontaneously hypertensive rats (SHR). Endothelium-dependent relaxation to sub-maximal acetylcholine (ACh) was greater in 16wk SHR vs. WKY. Maximal ACh caused contraction in 16wk and 30wk M, but not F. Age-related decrements occurred most notably in F-SHR, but not F-WKY. Indomethacin (COX inhibitor) restored relaxation to maximal ACh in 16wk M-SHR, and in 30wk M-WKY and M-SHR. Contraction to KCl was reduced in 16wk F-WKY and greater in 30wk M-SHR. Greater contractions to phenylephrine (PE) occurred in M vs. F (EC50, max. contraction). While 16wk rats were more sensitive (EC50), 30wk rats had greater maximal PE responses. 16wk rats had elevated aortic protein levels of the prooxidant subunits gp91phox (in SHR vs. WKY) and p67phox (in F-SHR), but not p22phox. In contrast, similar enzyme levels occurred in all groups of 30wk rats. These data support hypertension- and male sex-related decrements in endothelial function in 16wk rats that are exacerbated with age. In addition, female sex-related preservation of endothelial function is lost by 30 weeks of age in this model of hypertension. Research funded by the Heart and Stroke Foundation of Ontario.
