Calcium ion influx through voltage-gated calcium channels is an essential step in action potential–triggered release of neurotransmitters from presynaptic terminals and, hence, in the transfer of information in the nervous system. In addition to their role as calcium ion entry points, these ion channels are subject to both positive and negative modulation via a number of molecular signaling pathways. These regulatory mechanisms are important to the control of synaptic strength and plasticity and as potential pharmaceutical targets. This chapter reviews the principal mechanisms of presynaptic calcium channel regulation via pathways that include G-protein–coupled receptors, phosphorylation by protein kinase C, and their interaction with companion elements of the transmitter release site.
