Detection and molecular characterization of an aster yellows phytoplasma in common groundsel (Senecio vulgaris L.) in Canada

Abstract
A phytoplasma-induced yellows disease in common groundsel (Senecio vulgaris L.) was observed at an experimental farm near Brooks, Alberta, in 1996. Symptoms included reddening, chlorosis, plant stunting, proliferation of axillary shoots, and malformations (virescence and phyllody) of floral parts. Transmission electron microscopy revealed phytoplasma bodies in sieve tube elements of diseased plants, but not in healthy plants. These bodies were oval to spherical, lacked cell walls and were surrounded by single membranes. Their diameters ranged from 0.32-0.60 x 0.28-0.56 pm and averaged 0.44 x 0.38 μm. Using the polymerase chain reaction (PCR) and a pair of universal primer sequences, R16F2/R2, derived from phytoplasma 16S rRNA, an amplified product of the expected size (1.2 kb) was observed in samples from infected groundsel, but not in asymptomatic plants. Sequence analysis of the PCR products produced by the 16S-23S rDNA intergenic spacer region (SR) primers SRI and SR2 indicated that groundsel phytoplasma and aster yellow phytoplasma are closely related genetically. This is the first detailed report of a phytoplasma disease of common groundsel in Canada.
