Hepatocyte proliferation and differentiation are greatly affected by extracellular matrix (ECM). Integrin‐linked Kinase (ILK) is a cell‐ECM‐adhesions component implicated in cell‐ECM signaling via integrins. We investigated the role of ILK in liver size regulation in vivo by utilizing the LoxP/Cre model system. Targeted deletion of ILK in mouse liver (ILK‐KO) resulted in histological abnormalities in the liver characterized by disorderly hepatic plates, increased hepatocyte proliferation and apoptosis, increased proliferation of biliary cells, and increased deposition of extracellular matrix soon after birth. Microarray analysis indicated upregulation of integrin and matrix‐related genes and a concurrent downregulation of differentiation‐related genes. Western blot analysis indicated change in the expression of many integrins in the liver including integrin β1, β3, β5, and αV. Furthermore, The liver‐targeted ILK‐KO mice had increased liver‐to body‐weight ratio and fail to return to normal liver weight following partial hepatectomy. This increase in liver size was due to prolonged cell proliferation observed in the ILK‐KO mice even at 14 days after partial hepatectomy. Our results highlight the significance of ILK and hepatic ECM‐signaling for hepatocyte proliferation and liver size regulation.
