Abstract

As a monogenic recessive trait, hemophilia represents an ideal candidate for the application of somatic cell-based gene therapy. While the process of gene therapy is conceptually straightforward, intensive investigation over the past two decades has indicated that effective and safe gene transfer approaches are challenging to develop. This chapter reviews the basic components required to establish a successful gene transfer program. Current optimal approaches to gene therapy will be highlighted and the ongoing challenges to securing long-term clinical success of this therapeutic paradigm are summarized.
