Abstract
Ischemic stroke is initiated by the lack of oxygen and glucose that are normally involved in maintaining the ion gradients at the presynaptic terminals. Overstimulation of postsynaptic glutamate receptors induces a massive influx of calcium, mitochondrial depolarization, and accumulation of reactive oxygen species causing neuronal death. To begin to understand the cellular effects on neurons following a stroke, we hypothesized that protein ubiquitylation would be altered due to its crucial role in cell signalling and protein labeling for subsequent degradation. To test this hypothesis, we analyzed immunoblots of glutamate‐treated primary rat cortical neuron cultures. We found that several low‐molecular weight proteins in the glutamate‐treated neuron lysates showed increased ubiquitylation levels as compared to untreated neuron lysates. The altered ubiquitylation profile of only a subset of proteins suggests that select small proteins are tightly regulated during stroke. Through this study, we have identified proteins specifically targeted during stroke. Their identification opens the door to targets for therapeutic intervention to rescue neurons following ischemic brain injury. Grant Funding Source : This study was funded through Departmental Funds
