Absorption of sugarcane policosanols in golden syrian hamsters

Abstract
Supplementation with Cuban sugarcane policosanols (PC) has been shown to reduce total cholesterol and LDL‐C up to 23 and 31%, respectively. PC ability to decrease cholesterol levels stems from evidence showing that PC inhibit cholesterol biosynthesis. Octacosanol (C28) is believed to be the primary PC responsible for efficacy since the concentration of C28 is markedly larger compared to other PC found within the supplement. Nonetheless, intestinal absorption of PC could be limited by their hydrophobic nature, secondary to their aliphatic, saturated molecular structure. The objective of the present study was to evaluate levels C28 in the small intestine, liver, perirenal adipose, plasma and feces of male Golden Syrian Hamsters (N=36) fed diet containing no PC or 0.275 mg/g of either Cuban sugarcane PC or non‐Cuban sugarcane PC. A subset of 6 hamsters receiving non‐Cuban PC was fed 2.75 mg PC/g of diet. Results indicated that C28 was undetectable in tissues and plasma of animals supplemented with PC regardless of the brand or dosage. Fecal analysis revealed C28 was excreted 43% (P = 0.01) faster in animals receiving Cuban PC compared to those fed 0.275 mg PC/g non‐Cuban PC. Furthermore, C28 excretion was 38% (P = 0.05) faster in the Cuban PC group compared to the non‐Cuban group receiving 2.75 mg PC/d of diet. The results from this study suggest that insufficient amounts of supplemental PC are absorbed to elicit an effect on endogenous cholesterol biosynthesis.
