Sodium and glucose co‐transporter SGLT1 protein expression is regulated by eukaryotic protein synthetic initiation and elongation factors in the formula‐fed neonatal pig

Abstract
Our previously observed disassociation between SGLT1 protein and mRNA expressions prompted us to examine how SGLT1 protein expression is regulated at the translation level along the jejunal crypt‐villus axis in fed neonatal pigs. Six Yorkshire gilts were removed from sows at the age of d 5 and fed a commercial source of liquid formula till 14–16 d of age before being sacrificed for tissue collections. Three jejunal epithelial cell fractions, representing cells from the upper villus, the middle villus and the crypt regions, were sequentially isolated (cell viability of 92–95%) by the distended sac method. Western blots were used to examine eukaryotic protein synthetic initiation and elongation factor expressions. A higher (P<0.05) total eukaryotic initiation factor 4E (eIF4E) protein and higher phosphorylated‐eIE4E binding protein (γ‐form) were observed in the crypt cells. No differences (P>0.05) were observed in total eukaryotic elongation factor 2 (eEF2) protein abundance among the upper, the middle and the crypt cell fractions but lower (P<0.05) phosphorylated‐(Pi)‐eEF2 protein level and the Pi‐eEF2 to total eEF2 ratio were observed in the crypt cells. These results suggest that the abundant SGLT1 protein expression in the neonatal gut crypt cell is due to their higher cellular protein synthetic capacity in the formula‐fed neonatal pigs.
