Dipolar-Based Torsion Angle Measurements for Protein Structure Determination

Abstract
Torsion angles encode information on the backbone secondary structure and on the conformation of the side chains of a protein. Solid-state nuclear magnetic resonance (NMR) relies on the orientation dependence of nuclear spin interactions and can be used to determine torsion angles. This article describes various techniques developed in the last decade for applications in proteins under magic-angle spinning (MAS) conditions. We discuss both the techniques suitable for applications in proteins with selectively introduced nuclear spin labels and multidimensional dipolar chemical shift correlation methods permitting simultaneous site-specific measurements of a large number of torsional restraints.
