Glutaraldehyde fixed porcine aortic valves are commonly used for the construction of bioprostheses, yet little is known about the function of the two major structural components of the valve leaflet (the fibrosa and ventricularis), or how each is individually affected by the fixation process. We therefore examined the function of the fibrosa and the ventricularis by separating them from the aortic valve leaflet and testing them individually. These mechanical tests showed the normal fibrosa and ventricularis to have very different elastic moduli and extensibilities. Aortic leaflet extensibility in the radial direction occurs mainly from the unfolding of the corrugatedfibrosa, whose folds are maintained by its attachment to the ventricularis. In contrast, the leaflets of glutaraldehyde fixed valves showed similar extensibilities and elastic moduli for the two layers. Such alterations in the function and interactions of valve leaflet components following fixation may contribute to the poor durability of glutaraldehyde fixed xenografts.
