Abstract
Diante da ausncia de tratamentos eficazes para a esclerose mltipla (EM) e sabendo que peptdeos derivados de venenos animais tm sido usados como prottipos farmacolgicos para o desenvolvimento de novas drogas avaliamos o efeito do Tnp, um peptdeo cclico indito e com potencial antiinflamatrio derivado do veneno do peixe peonhento Thalassophryne nattereri, na encefalomielite autoimune experimental (EAE), um modelo representativo da EM. Avaliamos tambm a participao da citocina IL-10 na modulao dos efeitos do Tnp. Para isto, induzimos a EAE ativamente em camundongos C57BL/6 fmeas WT ou IL-10 KO pela imunizao com o MOG 35-55 , um peptdeo imunodominante da mielina. Demonstramos que a aplicao subcutnea do Tnp (3 mg/Kg) em vrios esquemas de tratamento: profiltico (0-9 dias), teraputico (10-19 dias) ou contnuo (0-19 dias) consegue diminuir a intensidade dos sintomas clnicos e adiar o pico de aparecimento dos sintomas graves por mecanismos tambm dependentes de IL-10. O uso de histologia, zimografia e citometria de fluxo, nos permitiu observar que o Tnp beneficamente interfere no circuito imunolgico da EAE em vrios estgios: 1) suprime
