Spatial and temporal expression of microsomal glutathione <i>S</i> ‐transferase‐1 in the female rat reproductive tract

Abstract
The microsomal glutathione S ‐transferases (MGSTs) are thought to play a cytoprotective role under conditions of oxidative or nitrosative stress. Our objective was to determine the site(s) of subcellular localization of protein and gene expression of MGST‐1 in the rat female reproductive tract. Immunoblot analysis revealed the presence of MGST‐1 (≈ 17 kDa) and in situ hybridization indicated the presence of MGST‐1 mRNA transcripts in the ovary, oviduct and uterus. LM immunohistochemistry demonstrated intense immunoreactivity for MGST‐1 in granulosa lutein cells in the ovary, and moderate immunoreactivity in the oviduct and uterus. Interestingly, whereas protein expression of MGST‐1 was detected mainly in the granulosa lutein cells, its mRNA transcripts were found in both the corpus luteum and ovarian follicles. Laser capture microdissection was used in conjunction with real time RT‐PCR to quantitatively compare the level of MGST‐1 gene expression in the granulosa and granulosa lutein cells. Consistent with the immunohistochemistry data, the level of MGST‐1 gene expression was 2.6‐fold higher in granulosa lutein cells of the corpus luteum relative to granulosa cells of the ovarian follicle. These results suggest that MGST‐1 may play an important role in protecting granulosa lutein cells against reactive oxygen or reactive nitrogen species. (Supported by CIHR)
