60, 120 and 240 min after administration of diclofenac, centrifuged and the supernatant stored at )80 °C until analysis.Serum concentrations of dicolfenac were measured by reverse phase high performance liquid chromatography with ¯uroimetric detection.Results There were no adverse effects following rectal administration of diclofenac 2 mgákg )1 and no incidence of primary or secondary bleeding.Individual serum drug concentration ®tted a biexpontential function and could be described in all patients.Pharmacokinetic parameters are expressed in Table 1 (mean SD).Discussion t max after rectal diclofenac varied from 30 to 60 min which is similar to that reported in adults (1).t 1/2B (71 20 min) values were similar to values after intravenous infusion administration in children ( 2).In this study, C max of 2.38 mgál )1 was recorded in comparison to a previous 1.6 mgál )1 report after 1.5 mgákg )1 in children after oral administration (3).C max is considerably lower than adult values possibly due to difference in V d .Rectal dicofenac pharmacokinetics mirrors adult elimination values and time to maximum concentrations.Diclofenac in children is rapidly absorbed after rectal administration and may be administered at induction of anaesthesia to provided adequate postoperative analgesia in children undergoing TA surgery.
