Abstract
The data reviewed support the hypothesis that the hippocampus is a key region in the pathophysiology of depression.Studies over the past decade suggest that a stress-related reduction of BDNF could contribute to depression and that MAMMs mediate their therapeutic benefit, in part, by increasing levels of this factor in the hippocampus.The regulation of other factors, such as VEGF or IGF-I may also be important.Finally, these and other factors may determine how neurogenesis is regulated in the hippocampus and link stress to decreased rates of neurogenesis, with MMAM treatment leading to, and perhaps relying on, increased rates of neurogenesis.Many questions remain, but with convergent evidence from clinical and molecular studies, provocative models of stress, depression and antidepressant mechanisms are emerging.
