Evaluation of an Enzyme Linked Immunosorbent Assay (ELISA) for Determination of Plasma Thyroxine in Dogs

Abstract
Summary Plasma thyroxine (T4) content, pre- and post-TSH stimulation, in euthyroid and primary hypothyroid dogs was simultaneously determined by an enzyme linked immunosorbent assay (ELISA) and a radioimmunoassay (RIA). Using the ELISA technique no significant difference in resting T4 levels between euthyroid and hypothyroid dogs was found. However, after TSH stimulation the increase in plasma thyroxine in euthyroid dogs was significant and greatly exceeded the T4 elevation in primary hypothyroid dogs (p ≦ 0.001). Consequently the ELISA technique can be used to confirm suspected primary hypothyroidism in dogs. Zusammenfassung Auswertung einer enzymatischen Immunomethode (ELISA) zur Bestimmung von Plasmatyroxin bei Hunden Die Konzentration von Plasmatyroxin vor und nach TSH-Stimulierung von eutyroiden und primär hypotyroiden Hunden wurde mit einer enzymatischen Immunomethode (ELISA) und einer radioimmunologischen Methode (RIA) bestimmt. Bei der Anwendung von ELISA wurde kein signifikanter Unterschied zwischen dem basalen T4-Spiegel von eutyroiden und hypotyroiden Hunden gefunden. Nach TSH-Stimulierung war jedoch die Steigerung des Plasmatyroxins bei eutyroiden Hunden signifikant und übertraf diejenige bei primär hypotyroiden Hunden bedeutend (p ≦ 0.001). Folglich kann die ELISA-Technik für die Feststellung von mutmaßlichem primären Hypotyroidismus bei Hunden angewendet werden. Toutefois, après la stimulation TSH, l'augmentation de thyroxine plasmatique chez les chiens euthyroïdiens était significative et dépassait fortement l'élévation T4 chez les chiens présentant une hypothyroïdie primaire (p ≦ 0,001). En conséquence, la technique ELISA peut être appliquée pour confirmer un cas suspect d'hypothyroïdie primaire chez les chiens. Con ELISA no se detectaron diferencias en los niveles de T4 de ambos grupos de animales antes de la estimulación con TSH. En cambio, después de la estimulación el aumento de T4 en los eutiroideos fué significativo y superó con amplitud (p < 0,001) los niveles de T4 de los perros con hipotiroidismo primario. Se concluye que el método ELISA puede ser utilizado para confirmar el hipotiroidismo primario en el perro.
