P4‐119: Crystal Structures of Apo Centaurin Alpha1 and its Complex With Phosphatidylinositol 3,4,5‐triphosphate

Abstract
Centaurin alpha1 (CENTA1) is an adaptor protein for the transport of phosphatidylinositol 3,4,5-triphosphate vesicles (PIP3ves) by kinesin motor KIF13B and involved in axonal development. The expression of CENTA1 is elevated in the neurons of Alzheimer's patients and part of neuritic plaques x-ray crystallography we solved the crystal structure of CENTA at 1.9Å resolution. The protein contains three domains, the N-terminal ArfGAP and the tandem PH domains. The PIP3-bound structure was also solved at 1.7Å resolution. The head group of PIP3 forms polar interactions with similar residues of both PH domains whereas the diacylglycerol group is not visible. CENTA1 is known to interact with nucleolin, involved in Alzheimer's disease pathology and tends to be phosphorylated in patients with Alzheimer's disease. The structures of CENTA1 are basis for understanding its interaction with nucleolin in molecular details and key for finding its association with Alzheimer's disease.
