A rapid quantitative PCR (qPCR)-based method for the detection of ale yeast (Saccharomyces cerevisiae) in rinse water was developed as a means of assessing vessel hygiene. Yeast cells were collected from spiked samples using a novel hollow-fiber filtration unit (ElutraSep module) that allows cell recovery by elution in a small final volume. The efficiency of the filtration device was determined by analysis of the growth of recovered cells on solid medium and compared with standard membrane filtration. In conjunction, cells were detected using qPCR with primers specific to S. cerevisiae. The presence of yeast was indicated by DNA amplification (cycle threshold value) and confirmed by melting-point analysis. DNA could be identified in samples containing between 5 × 102 and 5 × 103 total cells. Given that volumes of up to 15 L can be filtered through the ElutraSep device, this corresponds to a potential detection limit of less than 1 cell in 3 mL of rinse water. The method described here allows the detection and quantification of yeast in rinse-water samples within a period of 2–4 hr, without the need for preenrichment. Consequently, a rapid evaluation of yeast vessel hygiene can be achieved, leading to potentially faster response times.
