Zusammenfassung Das Hämolysin des Rauschbrand-Bazillus ist ein echtes, lösliches, spezifisches, bakterielles Toxin (Hämotoxin), welches verhältnismäßig empfindlich ist gegenüber äußeren Einflüssen. Durch Anwesenheit von mit Pepsin verdautem, hitzesterilisiertem Pferdeserum im Nährboden wird seine Bildung gehemmt und durch Formalineinwirkung verliert es seine toxischen (hämolysierenden), nicht aber seine antigenen Eigenschaften. Nach Injektion dieses Toxoids (Hämotoxoids) treten im Blutserum von Tieren spezifische Antihämolysine auf, die in vitro im Neutralisationstest nachgewiesen werden können. Diese Antihämolysine werden von der Mutter auf das neugeborene Kalb ausschließlich durch die Kolostralmilch derart übertragen, daß das Kalb annähernd einen gleich hohen Antihämolysin-Titer wie die Mutter aufweist. Das Hämotoxoid kann sich mit seinen spezifischen Antihämolysinen verbinden und das Antihämolysin-Bindungsvermögen einer Rauschbrand-Vakzine geht mengenmäßig offenbar parallel mit der immunisierenden Wirksamkeit dieses Impfstoffes. Diese einfach und exakt durchzuführende in-vitro-Methode bietet deshalb ein geeignetes Hilfsmittel zur Wertbemessung von Rausch-brand-Impfstoffen. Auch das Hämotoxoid scheint gegenüber einer längeren Lagerung empfindlich zu sein, weil keimfreie (Filtrat-) Impfstoffe verhältnismäßig frühzeitig sowohl ihr Antihämolysin-Bindungsvermögen als auch ihre immunisierende Wirksamkeit verlieren. Daß dies bei abgetöteten Vollkulturen (Anakulturen) nicht in dem Maße der Fall ist, wird damit erklärt, daß durch autolytische Vorgänge während der Lagerung ständig neues Antigen frei und der Verlust dadurch ausgeglichen wird. An Experiment in Evaluating Blackleg Vaccine in vitro The haemolysin of Cl. chauvoei is a true, soluble, specific bacterial toxin (haemotoxin) which is comparatively sensitive to environmental influences. The presence of pepsin-digested, heat-sterilized horse-serum in the culture medium hinders its formation. The addition of formalin destroys its toxic properties without affecting the antigenic activity. After injection of this haemotoxoid into animals, specific antihaemolysins develop in the serum as shown by the in vitro neutralisation test. These antihaemolysins are transmitted to the calf solely by the dam's colostrum, giving approximately the same titre in both. The haemotoxoid can combine with its specific antihaemolysin and the combining ability of a blackleg vaccine closely follows quantitatively the immunizing power of the vaccine. This simple and exact in-vitro method is therefore thought to represent a suitable method for evaluating blackleg vaccines. The haemotoxoid appears to be affected by long storage and there is a comparatively early loss in the antihaemolysin-combining and immunizing ability of sterile culture filtrates. This is less noticeable with formolised whole cultures (anacultures), possibly because autolytic processes progressively release new antigen during storage. Un essai pour éprouver in vitro la valeur des vaccins contre le charbon symptomatique L'hémolysine des bacilles du charbon symptomatique est une toxine bactérienne véritable (hémotoxine), soluble et spécifique et relativement sensible aux influences extérieures. La présence de sérum de cheval digéré par la pepsine et stérilisé à la chaleur dans les milieux de culture inhibe la production de la toxine, tandis que sous l'action du formol, elle perd ses propriétés toxiques (hémolysantes), mais non son caractère antigène. Après injection de cette anatoxine (hémotoxoïde), des antihémolysines spécifiques apparaissent dans le sérum sanguin des animaux et peuvent être décelées in vitro par le test de neutralisation. Ces antigènes sont transmis uniquement par le colostrum de la mère au veau nouveau-né si bien que le veau présente à peu près le même titre d'antihémolysines que la mère. L'hémotoxoïde (anatoxine) peut se combiner avec son antihémolysine spécifique et le degré par lequel un vaccin contre le charbon symptomatique est capable de fixer l'antihémolysine correspond quantitativement semble-t-il au pouvoir immunisant du vaccin. Cette méthode simple, pouvant être effectuée in vitro d'une façon exacte offre donc un moyen approprié d'éprouver la valeur des vaccins contre le charbon symptomatique. L'hémotoxoïde semble elle-aussi, être détruite par une conservation un peu longue, car les vaccins stériles (obtenus par filtration), perdent relativement tôt, aussi bien leur faculté de fixer l'antihémolysine que leur pouvoir immunisant. Le fait que cela ne se produise pas dans une si grande mesure avec les cultures tuées non filtrées (anacultures) peut être expliqué par des phénomènes autolytiques libérant constamment de nouveaux antigènes au cours de la conservation et compensant ainsi les pertes de ces éléments. Un ensayo para valorar in vitro las vacunas contra el carbunco sintomático La hemolisina del Clostridium feseri es una toxina (hemotoxina) bacterina verdadera, soluble y específica, que es relativamente labil frente a los agentes externos. Su formación se inhibe con la presencia en el medio nutritivo de suero sanguíneo de caballo esterilizado al calor y digerido con pepsina, y mediante la actuación del formol pierde sus propiedades tóxicas (hemolíticas), pero no las antigénicas. Tras la inyección de esta anatoxina (hemoanatoxina) aparecen antihemolisinas específicas en el suero sanguíneo de los animales, que se pudieron demostrar in vitro con la prueba de neutralización. Estas antihemolisinas se transmiten exclusivamente de la madre a la ternera recién nacida por medio de la leche colostral, de manera que la ternera presenta un título de antihemolisinas aproximadamente tan alto como el de la madre. La hemoanatoxina se puede combinar con sus antihemolisinas específicas y la capacidad de combinación con las antihemolisinas de una vacuna contra el carbunco sintomático está ligada, al parecer, cuantitativamente a la actividad inmunizante de esta vacuna. Esta técnica que se puede realizar in vitro con facilidad y exactitud representa por esta razón un medio auxiliar apropiado para la valoración de las vacunas contra el carbunco sintomático. También parece ser que la hemoanatoxina es sensible a un almacenamiento prolongado, puesto que las vacunas estériles (filtradas) pierden relativamente pronto tanto su capacidad de combinación con sus antihemolisinas como su actividad inmunizante. El que la pérdida no sea tan grande en los cultivos totales muertos (anacultivos) se explica por el hecho de que mediante los procesos autolíticos durante el almacenamiento se libera constantemente antígeno nuevo y con ello se equilibra su pérdida.
