Effect of Prostaglandin-E <sub>2</sub> on Gastric Secretion and on Gastric Circulation of Totally Isolated ex vivo Canine Stomach

Totally isolated canine stomachs, perfused with homologous blood, were used for the study of the effect of prostaglandin-E&lt;sub&gt;2&lt;/sub&gt; (PGE&lt;sub&gt;2&lt;/sub&gt;) on gastric secretion and circulation. Ex vivo stomachs were stimulated by a constant infusion of histamine into the gastric artery. When histamine-induced secretion was stabilized, PGE&lt;sub&gt;2&lt;/sub&gt; was introduced intra-arterially for 1 h. Doses of PGE&lt;sub&gt;2 &lt;/sub&gt;tested were 500, 250 and 125 µg/h. After 1 h PGE&lt;sub&gt;2&lt;/sub&gt; infusion was interrupted, but histamine stimulation continued. PGE&lt;sub&gt;2&lt;/sub&gt; significantly inhibited the output of gastric juice, HCl and pepsin and reduced the concentration of HCl. The effect of PGE&lt;sub&gt;2&lt;/sub&gt; was still very marked after it was discontinued. PGE&lt;sub&gt;2&lt;/sub&gt; had a potent vasodilator action on gastric circulation: it reduced the gastric peripheral resistance and the gastric arterial perfusion pressure. This hemodynamic action of PGE&lt;sub&gt;2&lt;/sub&gt; was present only during infusion of the inhibitor.
