Summary Lungs of calves which were treated for induction of pulmonary edema by hydrocortisone acetate, by immunization against the challenging P. hemolytica and by aerosol of parainfluenza 3 virus prior to challenge with P. hemolytica, were examined histologically, histochemically and by immunofluorescence against P. hemolytica. Alveolar macrophages were the main phagocytozing cells for P. hemolytica in lungs. Flooding alveolar edema which developed in calves treated to induce pulmonary edema and in one group of calves treated by hydrocortisone, interfered with phagocytosis by alveolar macrophages. Another group of calves treated by hydrocortisone developed foaming alveolar edema which did not interfere with phagocytosis of P. hemolytica. Polymorphonuclear leukocytes participated in phagocytosis in calves with hemagglutinating antibodies against P. hemolytica. An early bronchiolitis was present in all challenged calves after 4 hours due to the presence of P. hemolytica. Nearly all calves developed minute areas of atelectasis in the predilection sites of deposition of P. hemolytica after 4 hours. A profound alteration — extrusion of the esterase positive particles from the surface of alveolar epithelial cells — occured in the edematous pulmonary tissue of calves which were treated for induction of edema and by hydrocortisone. Flooding alveolar edema caused washing out of the alveolar lining film. One calf treated with an aerosol of histamine had an impaired clearance of P. hemolytica and developed an early fibrinous pneumonia. Zusammenfassung Mikroskopische Befunde an Lungen von Kälbern die mit einem Pasteurella haemolytica-Aerosol und zur Veränderung der pulmonalen „Clearance” behandelt worden war Vier experimentelle Gruppen von Kälbern wurden mit einem P. Haemolytica Aerosol nach der Induzierung des Lungenödems, nach der Aplikation des Hydrocortison Azetats, nach der Immunisierung mit P. Haemolytica und nach der Belastung mit einem Parainfluenza 3 Virus-Aerosol behandelt. Die Lungen wurden pathohistologisch, histochemisch und immunfluoreszenzmikroskopisch auf P. haemolytica untersucht. In den Lungen waren die Alveolarmakrophagen diejenigen Zellen, die bei P. haemolytica hauptsächlich die Phagozytose übernommen hatten. Ein flüssiges Alveolarödem, das bei Kälbern auftrat, die zur Induzierung eines Lungenödems behandelt worden waren, sowie bei einer Kälbergruppe, die Hydrocortison erhalten hatte, störte die Phagozytose durch Alveolarmakrophagen. Bei einer anderen Kälbergruppe, die mit Hydrocortison behandelt worden war, entwickelte sich ein schaumiges Lungenödem. Das schaumige Lungenödem beeinflusste die Phagozytose von P. haemolytica nicht. Polymorphkernige Leukozyten nahmen bei Kälbern mit hämagglutinierenden Antikörpern gegen P. haemolytica an der Phagozytose teil. Eine frühzeitige Bronchiolitis war bei allen belasteten Kälbern nach 4 Stunden vorhanden. Bei fast allen Kälbern zeigten sich nach 4 Stunden kleine atelektatische Herde in den bevorzugten Ablagerungsstellen von P. haemolytica. In ödematösem Lungengewebe von Kälbern, die zur Induzierung eines Ödems behandelt worden waren oder Hydrocortison erhalten hatten, fand eine weitgehende Änderungverdrängung der esterasepositiven Partikel von der Oberfläche der Alveolarzellen statt. Das flüssige Alveolarödem verursachte ein Auswaschen des die Alveolen begrenzenden Films. Eine frühzeitige fibrinöse Lungenentzündung entwickelte sich in einem Kalb, das mit aerosoliertem Histamin behandelt wurde und eine gestörte Phagozytose zeigte. Résumé Résultats microscopiques sur des poumons de veaux qui ont été traités avec un aérosol de Pasteurella haemolytica et modification de la «clearance» pulmonaire Des veaux ont été traités de quatre façons différentes soit afin de provoquer de l'oedeme pulmonaire, avec de l'acétate d'hydrocortisone, par immunization contre P. haemolytica et enfin un aerosol de virus Parainfluenza 3, avant le «challenge» avec P. haemolytica. Les poumous de ces animaux ont été examinés au point de vue histologique histochimique et au moyen de l'immunofluorescence contre P. haemolytica. Les macrophages alvéolaires des poumons furent les principales cellules phagocytant P. haemolytica. L'oedème pulmonaire sévère, qui apparut chez les veaux traités dans le but de provoquer cet oedème pulmonaire et dans un groupe traité a l'hydrocortisone, empêcha la phagocytose par les macrophages alvéolaires. Un oedème pulmonaire spumeux se développa dans un autre groupe de veaux traités à l'hydrocortisone. L'oedème spumeux n'a pas influencé la phagocytose de P. haemolytica. Des neutrophiles participerènt également à la phagocytose chez les veaux qui possédaient des anticorps contre P. haemolytica. On a constaté l'apparition d'une bronchiolite légère quatre heures seulement après l'exposition de «challenge» des veaux à P. haemolytica. De petits foyers d'atélectasie apparurent aussi chez presque tous les veaux dans le sites de dépots de prédilection de P. haemolytica. Par contre, un changement majeur s'est produit, soit la libération de particules estérase positive de la surface de l'épithelium alvéolaire des veaux traités dans le but de provoquer de l'oedème pulmonaire ou avec de l'hydrocortisone. L'oedème pulmonaire extensive provoqua le lavage du film recouvrant les alvéoles. Un veau traité avec un aerosol d'histamine, qui a pour propriété d'altérer l'élimination de P. haemolytica, contracta une pneumonie fibrineuse legère. Resumen Hallazgos microscópicos en pulmones de terneros que se habían tratado con un aerosol Pasteurella haemolytica y para modificar el aclaramiento pulmonar Cuatro grupos experimentales de terneros se trataron con un aerosol P. haemolytica tras la inducción de un edema pulmonar, tras la aplicación de acetato de hidrocortisona, tras la inmunización con P. haemolytica y tras el sobrecargo con un virus-aerosol parainfluenza. Los pulmones se examinaron histológica, histoquímica e inmunofluorescente-microscópicamente en cuanto a P. haemolytica. Eran los macrófagos alveolares en los pulmones las células que habían asumido sobre todo la fagocitosis con P. haemolytica. Un edema alveolar líquido que apareció en terneros que se habían tratado para inducir un edema pulmonar, así como en un grupo de terneros que recibieron hidrocortisona, perturbaba la fagocitosis por medio de macrófagos alveolares. En otro grupo de terneros que habían sido tratados con hidrocortisona se desarrolló un edema pulmonar espumoso. El edema pulmonar espumoso no influía sobre la fagocitosis de P. haemolytica. En terneros con anticuerpos hemoaglutinantes frente a P. haemolytica intervenían leucocitos polimorfonucleares en la fagocitosis. En todos los terneros sobrecargados se hallaba presente tras 4 horas una bronquiolitis precoz referida a las especialidades de P. haemolytica. En casi todos los terneros se presentaban tras 4 horas unos focos atelectáticos pequeños en los lugares predilectos de depósito de P. haemolytica. En el tejido edematoso pulmonar de los terneros que habían sido tratados para inducir un edema y que habían recibido hidrocortisona, se realizó un desalojamiento amplio de cambio de las partículas esterasapositivas de la superficie de las células alveolares. El edema alveolar líquido ocasionaba un lavamiento de la película que limitaba los alveolos. Una pulmonía fibrinosa precoz se desarrolló en un ternero, el cual había sido tratado con histamina aerosolizada y que presentaba una fagocitosis turbada.
