Abstract Attempts to substantially increase the relative proportion of β-lactose octaacetate in products of acetylation of α-lactose monohydrate using various acetylation protocols failed. Nevertheless, the revised protocol for isolation and crystallization, based on the classical work by Hudson and Johnson, rendered preparation of β-lactose octaacetate less labor intensive. It is proposed that the melting point and [α]D value are not reliable criteria of purity of β-lactose octaacetate, which can be confidently determined by 1H NMR spectroscopy. Notes The term lactose is a trivial name reserved[ Citation 1 ] for β-D-galactopyranosyl-(1→4)-α,β-D-glucopyranose (or 4-O-β-D-galactopyranosyl-α,β-D-glucopyranose). The terms D-lactose or D-lactopyranose, which can often be found in print, are notoriously incorrect. The "D" is redundant because of the definition, and "…pyran…" is obvious because of the (1→4)-linkage involved in the structure and, therefore, unnecessary. Sigma-Aldrich has discontinued offering the compound (>98% purity), and the current price by the sole supplier in the United States, 3B Scientific Corporation (www.3bsc.com), is $980/100 g. Toronto Research Chemical, Inc. lists the compound at $1000/100 g, but the anomeric purity of the material is uncertain.
