P1110 Background: Immunosuppressive effects of Baohuoside-1 (B1), a novel flavonoid isolated from Epimedium, were evaluated in vitro and in vivo in this study. Methods: Proliferation assay was used for determine the antiproliferative prosperities. Flow cytometry analysis was applied to detect the changes of phenotypes on activated cells. Analysis of Western blotting was employed to observe the expression of cyclins and cdks proteins. Apoptosis induction was analysed by annexin V/PI staining. Levels of IL-2 and IL-2R proteins were evaluated by ELISA and FACS. A model of rat heart transplantation was used to further determine the immunosuppression of B1 in vivo. Results: The major findings are as follows: (1) B1 has immunosuppressive effect on MLC with IC50 of a low μM concentration. (2) B1 effectively inhibits the proliferation of human T and B cells triggered by IL-2 or mitogenic antigen stimulation in a dose and time dependent fashion. (3) B1 resulted in G1−S phase cell arrest. (4) B1 down-regulated the expression of cyclin A and p33cdk2 proteins on IL-2 signalling, but had only marginal effect on expression of cyclin D protein. (5) B1 could induce apoptosis and caspase-3 activation in human T lymphocytes. (6) B1 declined partially the level of secretion of IL-2 protein and expression of IL-2R. (7) The synergistic immunosuppressive effect between B1 and FK506 was found in the CD69 expression stimulated by PMA/CD28 and PMA/iono, but not by CD3/CD28. (8) Furthermore, synergistic effect of B1 combined with FK506 was also observed by prolonging MST in rat heart transplant model in vivo. Conclusions: The immunosuppression of B1 on cell activation takes place in G1−S transition and is associated with expression of IL-2 and IL-2R, apoptosis induction and with expression of cyclin A and p33cdk2 proteins. B1 has selectively immunosuppressive effects on T and B cell activation in vitro and in vivo.
