Summary The separation of mouse spermatogenic cell nuclei by sedimentation velocity at unit gravity has been used to determine the timing of histone and “mouse protamine” synthesis, and the turnover of basic nuclear proteins throughout spermatogenesis. Animals were injected with 3H-arginine or 3H-lysine and at various time intervals (2 hours post-label or from 1 to 30 days post-label) germinal cell nuclei preparations were separated on the staput. Labelled histories and mouse protamine were extracted from staput separated nuclei with hydrochloric acid and fractionated by polyacrylamide gel electrophoresis. Results indicate that histones are synthesized in association with DNA replication in spermatogonia and preleptotene spermatocytes, in pachytene primary spermatocytes and in spermatids stages 11 - 16, simultaneously with “mouse protamine”. Experiments are reported showing that histones synthesized in pachytene primary spermatocytes and in spermatids stages 11–16 are retained in epididymal spermatozoa, while histones synthesized before meiosis are no longer detectable onto chromatin after meiosis. Zusammenfassung Zur Markierung von Histonen und “Mäuse” -Protaminen erhielten adulte Mäuse intratestikulär 3H-Arginin oder 3H-Lysin. Nach verschiedenen Zeitintervallen (2 Std nach der Markierung oder 1 Tag bis zu 30 Tage nach der Markierung) wurden mit Hilfe der “Staput-Technik” fünf Zellkernfraktionen des Samenepithels gewonnen, welche sich aus Kernen ganz bestimmter Zelltypen zusammensetzten. Die markierten Histone und “Mäuse”-Protamine wurden mit HCl extrahiert und durch Polyacrylamidgel-Elek-trophorese fraktioniert. Dadurch war es möglich, diejenigen Zelltypen und deren Phasen zu erfassen, in denen die Synthese der Histone und “Mäuse”-Protamine stattfindet sowie das Turnover der basischen Kernproteine während der Spermatogenese zu bestimmen. Die Ergebnisse zeigen, daß die prämeiotische Histonsynthese der Spermatogonien und präleptotänen Spermatocyten während der DNS-Synthese stattfindet, die Histonsynthese in der meiotischen Prophase während des Pachytänstadiums der Spermatocyten und die Histonsynthese nach der Meiose während der Stadien 11 - 16 in den Spermatiden, in welchen auch die Synthese der Mäuseprotamine stattfindet. Es wird ferner darauf hingewiesen, daß die prämeiotisch synthetisierten Histone entweder während der Meiose oder der nachfolgenden frühen Spermiogenese ihre Affinität zum Chromatin verlieren und daher nicht mehr nachgewiesen werden können, während die in der meiotischen Prophase und danach gebildeten Histone noch in epididymalen Spermatozoen dargestellt werden können. Resumen Para determinar el tiempo de síntesis de histona y protamina, y el metabolismo de proteinas nucleares básicas a través de la espermatogénesis, se nan utilizado nucleos de células espermatogénicas de raton separadas por velocidad de sedimentación. A los animales se les inyecto argimina-H3 ó lisina-H3, y a distintos intervalos (2 horas post-marcaje, ó de 1 a 30 dias después del mismo). Se separaron los núcleos de células germinales. Los histonas y protaminas marcadas de ratón se extrajeron de los núcleos, separados con ácido hidroclórico y fraccionados mediante eletroforesis en gel de poliacrilamida. Los resultados indican que los histonas se sintetizan en asociación con la replicación del DNA en la erspermatogoniay espermatocitos preleptotene, en espermatocitosprimarios paquitene y en espermátides, estadios 11–16, simultaneamente con la “protamina del ratón” se aportan experimentos que muestran que las histonas sintetizadas en espermatocitos primarios paquitenicos y en espermátides, estadios 11 - 16, se conservan en los espermatozoides epididimarios, mientras que lashistonas sintetizadas antes de lameiosis no son detectables sobre la cromatina después de la meiosis.
