Kompartment-spezifische Expression von TGF-β abhängigen Genen in einem Emphysem-Tiermodell

Abstract
Transforming growth factor-β (TGF-β) spielt eine zentrale Rolle für die Homöostase des Bindegewebes in der Lunge. Die Entwicklung eines Emphysems ist mit der verringerten Expression von TGF-β sowie seiner Rezeptoren und Signalmoleküle assoziiert. In dieser Studie wurde die kompartmentspezifische Expression von Wachstumsfaktoren, tissue inhibitor of matrix proteinase-1 (TIMP-1) und Kollagen-1 in Smad3 -/- Mäusen untersucht, die spontan ein Lungenemphysem entwickeln. Es wurden 5 Smad3 -/- Mäuse sowie 8 korrespondierende Wildtypen (Smad3 +/+ ) zu den Zeitpunkten 12 Wochen und 20 Wochen untersucht. Die kompartmentspezifische Genexpression wurde mittels Laser-capture Mikrodissektion sowie nachfolgender quantitativer Real-time-PCR untersucht. In den Atemwegen von Smad3 -/- Mäusen zeigte sich eine signifikante Reduktion der Expression von TGF-β1 (12 Wochen: 0.44±0.14), TGF-β3 (20 Wochen: 0.094±0.048), connective tissue growth factor (CTGF) (12 Wochen: 0.37±0.063) und Kollagen-1 (20 Wochen: 0.36±0.087) (jeweils Mittelwert±SEM der n-fachen Expression von Smad3 +/+ Mäusen, jedes P<0.05). In Pulmonalarterien wurde eine relevante Abnahme der Expression von CTGF (12 und 20 Wochen: 0.22±0.13 bzw. 0.33±0.098), vascular endothelial growth factor (VEGF) (20 Wochen: 0.40±0.15) sowie Kollagen-1 (12 und 20 Wochen: 0.53±0.10 bzw. 0.36±0.13) festgestellt (jeweils P<0.05 gegen Smad3 +/+ Mäuse). Weiterhin konnte ein signifikanter Rückgang der Transskription von CTGF (20 Wochen: 0.25±0.13), VEGF (12 und 20 Wochen: 0.54±0.070 bzw. 0.57±0.17) und Kollagen-1 (20 Wochen: 0.44±0.17) im alveolären Kompartment gemessen werden (jedes P<0.05 gegen Smad3 +/+ Mäuse). Die Ergebnisse dieser Studie zeigen, dass eine Reihe wichtiger Gene stromabwärts von TGF-β in allen 3 Kompartimenten bei Smad3 -/- Mäusen herabreguliert sind. Es kann postuliert werden, dass das Unvermögen, diese Gene nach mikroskopischen Gewebeschäden heraufzuregulieren, zu gestörten Reparationsvorgängen und schließlich zum Emphysem führen.
