Effects of aldosterone antagonism on structural and mechanical properties of small arteries altered by angiotensin II

Abstract
The renin-angiotensin-aldosterone system is responsible for many vascular changes observed in hypertension, contributing to increased morbidity and mortality. In the present study we evaluated the effect of spironolactone, an aldosterone antagonist, in preventing some of the vascular alterations caused by angiotensin II (Ang II). Four groups of male SD rats were studied, including SHAM (vehicle infusion), Ang II-infused (120 ng/Kg/min, SC), Ang II infused + Spironolactone (SPI 25 mg/Kg/d in food) and Aldosterone-infused (ALDO 750 ng/h, SC) + SPI rats. Systolic blood pressure was monitored by the tail-cuff method. Third-order branches of mesenteric arteries (lumen diameter < 300 μm) were mounted on a pressurized myograph and exposed to intraluminal pressures ranging from 3 to 140 mmHg. Media thickness and lumen diameter measured at each pressure level allowed to determine wall mechanical properties. At 45 mmHg and compared to Ang II infusion, spironolactone prevented partially the increase of media thickness (16.1 ± 0.4 vs 18.9 ± 0.4, p<0.001), reduced media-to-lumen ratio (7.1 ± 0.2 vs 8.1 ± 0.2 %, p<0.01) and media cross sectional area (12470 ± 810 vs 15240 ± 667 mm2, p<0.01) of mesenteric arteries. Stress-strain relationship was shifted to the left by Ang II and was superimposable with control in Ang II + spironolactone group. These findings suggest that some effects of Ang II on resistance arteries in rats may be mediated by aldosterone.
