Circulating lymphocytes obtained from normal adults have been fractionated by centrifugation in a discontinuous BSA gradient. Five fractions of lymphocytes were obtained. The bulk of the small lymphocyte population segregated in two fractions of intermediate density and in the pellet. Cells in all five of the fractions responded with blastogenesis and mitosis to stimulation with phytohemagglutinin. On the other hand, only four of the fractions responded to stimulation with ALS and three to stimulation with allogeneic cells. Only two fractions responded to stimulation with PPD. Investigations are aimed at demonstrating whether the cells in each active fraction can be further fractionated to yield cells capable of responding to stimulation with only one or two of the mitogenic agents used.
