CAMP-DEPENDENT PROTEIN KINASE ACTIVATION OF HSP70 EXPRESSION FOLLOWING EXERCISE

Abstract
Myocardial expression of the inducible isoform of the 70kDa heat shock protein (Hsp70) has been directly linked to cardioprotection following ischemia-reperfusion injury. While exercise represents a physiologically relevant means to increase myocardial HSP70 content, the regulatory mechanisms underlying this response remain to be elucidated. Trimerization, DNA-binding and phosphorylation of a heat shock transcription factor (HSF1) are believed to be necessary steps in this process. In the present study, male Sprague-Dawley rats were run on a motorized treadmill (30 m/min at 2% grade for 60 min) with or without the cAMP-dependent protein kinase (protein kinase A – PKA) inhibitor N-[2-(-p-Bromocinnamylamino)ethyl]-5-isoquinolinesulfonamide.2HCl (H89 – 360 μg/kg body weight). Animals were sacrificed and hearts extirpated at 30 min or 24 hours post-exercise. Exercise alone resulted in HSF1 DNA-binding and increased hsp70 mRNA by 30 min (p < 0.05) and an elevated Hsp70 content (p < 0.05) in the myocardium at 24 hours post-exercise. In the presence of the PKA inhibitor, myocardial HSF1 DNA-binding was still evident following exercise, however, the typical increase in hsp70 mRNA and protein were not observed. These findings suggest that PKA is an important upstream regulator of Hsp70 associated cardioprotection and that it's action is downstream of HSF1 DNA-binding. (Funded by HSFO and NSERC)
