Abstract
930 canaux cationiques, la nature moleculaire des proteines responsables de ces conductances est demeuree longtemps obscure. Recemment, deux equipes (l’une japonaise et l’autre americaine) ont annonce le clonage d’un ADN complementaire (ADNc) d’une proteine ayant une haute homologie avec le domaine senseur de voltage des canaux ioniques dependants du voltage [5, 6]. L’equipe de Sasaki [5] a clone cette proteine, nommee mVSOP (membrane voltage sensor only protein), chez la souris. De leur cote, Ramsey et ses collaborateurs [6] sont arrives a la meme sequence chez l’hom-me et, par cons e q u e n t , ont nomme la proteine Hv1 (human v o l t a g e s e n s o r one). Dans ces deux etudes, la s e q u e n c e primaire de la proteine clonee est homologue au senseur d e v o l tage et ne c o n t i e n t a u c u n e autre region homologue a un pore de canal > Les canaux a protons actives par le voltage, ont ete identifies pour la premiere fois en 1982 dans les neurones d’escargots [1] et ont ete caracterises egalement dans les cellules epitheliales alveolaires, les macrophages, les granulocytes, les osteoclastes, les microglies [2] et, plus recemment, dans les fibroblastes cardiaques humains [3, 4].
