Abstract
Abstract Die Rolling‐Circle‐Amplifikation (RCA) ist ein isothermer enzymatischer Prozess, der durch bestimmte DNA‐Polymerasen vermittelt wird. Bei der RCA werden lange einzelsträngige (ss) DNA‐Moleküle mithilfe eines einzelnen DNA‐Primers an einer kurzen ringförmigen ssDNA‐Matrize synthetisiert. Die RCA hat sich als eine Standardamplifikationsmethode für die ultraempfindliche DNA‐Detektion in der Genomik und Diagnostik bewährt. Neuerdings ist es gelungen, die RCA als Verfahren für die Erzeugung von DNA‐Matrizen im großen Maßstab nutzbar zu machen. Die so erzeugten Matrizen können zur Herstellung von periodischen Nanoanordnungen eingesetzt werden. Darüber hinaus wurden verschiedene RCA‐Strategien entwickelt, um repetitive Sequenzen von DNA‐Aptameren und DNAzymen herzustellen, die als Detektionsplattformen für kleine Moleküle und Proteine fungieren. Diese neuen Verfahren machen die RCA zu einer höchst vielseitigen Methode für die Vervielfältigung von DNA, die breite Anwendungsmöglichkeiten in der Genomik, Proteomik, Diagnostik, Biosensorik, Wirkstoffentwicklung und Nanotechnologie finden wird.
