An Improved Method for the Isolation of Total RNA from Malva pusilla Tissues Infected with Colletotrichum gloeosporioides

Abstract
A modified RNA extraction method was developed for fungal and plant tissues that has several advantages compared with existing, published methods for materials with high contents of ribonuclease, phenolics and polysaccharides. The method uses guanidinium isothiocyanate in the homogenization buffer, and then hexadecyltrimethylammonium bromide (CTAB). soluble polyvinylpyrrolidone and insoluble polyvinylpolypyrrolidone in the extraction buffer. The procedure consistently produced a high yield of relatively high purity, undegraded RNA from a variety of biological materials and should be useful for other tissues where RNases, phenolics and sticky polysaccharides are a problem. Zusammenfassung Kin verbessertes Verfahren zur Isolierung der Gesamt-RNA aus Malva-pusilla-Geweben, die mit Colletotrichum gloeosporioides infiziert sind Fur pilzliche und pflanzliche Gewebe wurde ein modifiziertes Verfahren der RNA-Extraktion entwickelt, das im Vergleich zu anderen, bereits veroffentlichten Methoden verschiedene Vorteile bei Materialien mit hohem Gehalt an Ribonuclease. Phenolen und Polysacchariden bietet. Gearbeitet wird mit Guanidinisothiocyanat im Homogenisierungspuffer und CTAB, loslichem Polyvinylpyrrolidon und unloslichem Polyvinylpolypyrrolidon im Extraktionspuffer. Das Verfahren ermoglicht durchgehend hohe Ausbeuten an vergleichsweise sehr reiner nicht abgebauter RNA aus verschiedenen biologischen Materialien und ist sicherlich auch fur andere Gewebe nutzlich, bei denen RNasen. Phenole und klebrige Polysaccharide Probleme bereiten.
