Abstract
Chronic obstructive pulmonary disease (COPD) is associated with inflammation, irreversible airflow obstruction and muscular atrophy. Recently, it was shown that muscular atrophy in COPD is linked with alterations in satellite cell function which are the primary myogenic source contributing to skeletal muscle regeneration. The satellite cell‐marker CD34 plays a role in cell movement and muscle repair in toxin‐induced muscle injury. However, the link between CD34 expression and muscle regeneration in COPD has never been studied. Our hypothesis is that the inflammatory and hypoxic conditions found in COPD influence the expression of CD34 by satellite cells and affect muscle regeneration. The expression of CD34 was detected 1) in human satellite and C2C12 cell cultures in response to hypoxic or inflammatory conditions and 2) in muscle biopsies obtained from COPD or healthy subjects. Results show that CD34 expression in muscle decreases with COPD severity. Surprisingly, hypoxic and inflammatory conditions increased CD34 expression in cells, while myosin expression was reduced. This suggests that inflammatory and hypoxic stress increase CD34 expression in skeletal muscle cells which return to a satellite cell‐like phenotype. It also suggests an interesting role for CD34 in the response to hypoxic and inflammatory conditions, and a potential role for CD34 in muscle atrophy in COPD.
