Abstract
The W34 gene, which was first cloned into pGEM-T-EASY vector,was inserted into EcoRI site of pBDREB vector.A plasmid was selected, with which the bacteria host could grow on Amp plate; The plasmids pBW34-DREB、pSPROK、pBlueks were digested with the restriction enzymes PstI/KpnI、KpnI/EcoRI、PstI/EcoRI respectively, so three fragments were obtained, and then they were ligated with T4 DNA Ligase, thus we got the medium expression vector pBWD-Tnos;The vector pBWD-Tnos was cut with SmaI and EcoRV, a 2.16kb fracment including promotor and coding sequence of DREB was obtained.The fragment was then ligated to the vector pCDMAR-Hyg which had been cut with an enzyme SmaI, so the plant expression vector pCDMAR-pWDT-Hyg of the DREB protein encoding gene was finally obtained.The results of the restriction enzymes digestion demonstrated the correctness of the construction. The double T-DNA vector was transferred into the maize embryonic calli mediated by Agrobacterium tumefaciens. We hope we can obtain the transgene plants without selective marker genes.
