Introdução: o Retinoblastoma é o tumor intra-ocular maligno mais comum na infância. Pacientes com tumores avançados, em geral, respondem mal ao tratamento. Hipóxia é um fenômeno comum aos tumores avançados e está relacionada à resistência à quimio e radioterapia convencional. As células e os tecidos tumorais adaptam-se ao microambiente hipóxico através da ativação de várias vias e moléculas associadas à hipóxia. Destas, o fator induzido pela hipóxia 1 alfa (FIH-1-α) é o fator de transcrição mais importante que regula a resposta celular à hipóxia. Objetivo: avaliar a expressão de FIH-1-α no retinoblastoma e avaliar a proliferação das de células de retinoblastoma com o bloqueio do gen FIH-1-α. Material e Métodos: 21 casos de retinoblastoma foram selecionados do arquivo do The Henry C. Witelson Ocular Pathology Laboratory, Universidade McGill, Montreal, QC, Canadá. Foram coletados dados clínicos e realizado estudo histopatológico de todos casos, além de imunohistoquímica com o anticorpo FIH-1-α. A linhagem de célula de retinoblastoma humano Y79 foi cultivada. As células foram tratadas com Cloreto de Cobalto (CoCl2) com o intuito de estabilizar a proteína FIH-1-α. Após este tratamento, foi realizada análise dos níveis de RNAm de FIH-1-α por PCR e western blot. Foi realizado o método de siRNA para bloqueio do gen FIH-1-α e, após esse bloqueio, foi realizado ensaio de proliferação para avaliar o efeito na proliferação das células de retinoblastoma. Resultados: 85.7% dos casos de retinoblastoma expressaram FIH-1-α. Houve correlação entre a expressão de FIH-1-α com necrose e invasão vítrea. As células de retinoblastoma tiveram aumento significativo de RNAm de FIH-1-α após o tratamento com CoCl2. Após o bloqueio do gen FIH-1-α com siRNA, houve diminuição significativa da proliferação das células de retinoblastoma. Conclusão: Foi demonstrado que a maioria dos retinoblastomas expressam FIH-1-α, e que as células de retinoblastoma apresentam aumento de FIH-1-α após o tratamento com CoCl2. Além disto, o bloqueio do gen FIH-1-α levou a uma diminuição significativa na proliferação cellular. Assim, drogas que inibem FIH-1-α podem ser de grande utilidade no tratamento do retinoblastoma, principalmente em casos avançados onde existe muita hipóxia tumoral.
