Une mutation ponctuelle (N288D), associée à l'expression de l'hyperglycérolémie familiale, a été identifiée dans le gène de la glycérol kinase (GK) au sein de la population du Saguenay-Lac-Saint-Jean (SLSJ). La présente étude a comme objectif d'analyser les caractéristiques démogénétiques de proposants atteints de cette maladie et d'essayer de retracer les origines de la mutation responsable dans la région du SLSJ. Ainsi, à partir des reconstitutions généalogiques de 11 individus présentant cette mutation ainsi que de trois groupes témoins appariés, diverses mesures démogénétiques ont été effectuées et comparées. Les coefficients de consanguinité et d'apparentement sont, dans l'ensemble, supérieurs chez les proposants. L'analyse des fondateurs régionaux a permis de mettre en valeur l'importance des individus provenant de la région de Charlevoix et ce, dans les quatre groupes. Par ailleurs, une analyse approfondie des fondateurs régionaux a permis d'identifier un couple originaire de Baie-St-Paul qui aurait vraisemblablement introduit la mutation N288D dans la région du SLSJ. Il n'a toutefois pas été possible de cibler le ou les individus ayant introduit la mutation dans la population québécoise. Il s'agit de la première étude démogénétique ayant été réalisée sur une cohorte aussi nombreuse de cas d'hyperglycérolémie familiale.
