Co‐Activator of 150 kDa (CA150) is a transcription elongation factor, but has also recently been shown by our lab to act as a co‐repressor for CCAAT/enhancer binding protein α (C/EBPα) transcriptional activity. Interestingly, CA150 also inhibits the growth arrest activity of C/EBPα. In this study, we examined domains within CA150 that mediated its inhibitory activity towards C/EBPa and the mechanism whereby this effect occurs. Our structure/function analysis using mutants of CA150 suggest that the inhibitory activity lies within the N‐terminal domain. Additionally, laser‐scanning confocal microscopy was used to visualize the subnuclear localization of fluorescent‐tagged CA150 and C/EBPα. Results show that co‐expression of CA150 influences the intranuclear organization of C/EBPα, suggesting a possible mechanism whereby CA150 inhibits the growth arrest and transcriptional activities of C/EBPα.
