Abstract
以低氮(0.2 mmol/L)胁迫4 h的氮高效棉花品种新陆早12号为材料,提取棉花总RNA,根据蓖麻和杨树的 NRT 2 基因保守序列设计含酶切位点的特异引物,用RT-PCR技术获得350 bp的片段。将此片段以正向和反向插入到中间载体pUCM-T的GUS内含子序列两端。将正向片段-GUS-反向片段插入pPZP35S,通过酶切和PCR鉴定,成功构建RNAi载体,并成功转入农杆菌GV3101中,为研究棉花硝酸盐高亲和转运蛋基因 NRT 2 的功能奠定一定的基础。
