Abstract
为了获得具有多种抗逆特性的转基因甘薯材料，本文构建了由甘薯过氧化物胁迫诱导型启动子SWPA2和植物组成型启动子CaMV 35S分别控制、且含有甘薯SP8的结合蛋白基因( IbSPF1 )的过表达植物载体。IbSPF1是植物WRKY转录因子家族的成员之一。由于其参与植物多种生物和非生物胁迫的反应以及许多生理过程，WRKY类家族越来越成为人们研究的热点。本研究利用RT-PCR方法从甘薯茎叶组织中扩增出编码 IbSPF1 的完整开放阅读框，与已发表的 IbSPF1 相比，从翻译起始点开始第1 009位点的胸腺嘧啶转变为胞嘧啶，从而使翻译产物由色氨酸转变为脯氨酸。本研究进一步构建了由不同启动子控制、携带 IbSPF1 基因的超表达植物载体pCAMBIA2300，并转入到根癌农杆菌菌株EHA105中，为未来甘薯抗逆转基因打下基础。
