Abstract

Nearly 3 decades have elapsed since the initial characterization of native gonadotropin-releasing hormone. Numerous agonistic and antagonistic analogues have been synthesized in the interim and have been used to manipulate the menstrual cycle and treat various steroid-dependent disorders. The antagonists have also functioned as physiologic probes for the investigation of various menstrual cycle–related events. This article briefly reviews the development of agonists and antagonists, contrasts their mechanisms of action, and compares the clinical applications of the existing agonists with the recently developed antagonists.
