Abstract

牛乳のβ-ラクトグロブリンを免疫抗原として作製したモノクローナル抗体を用いて,特定原材料の通知法の抽出方法に対応した,新たな牛乳ELISAキットを構築した.(1)作製したモノクローナル抗体は,ドデシル硫酸ナトリウムと2-メルカプトエタノールで可溶化された牛乳標準品を検出可能であった.さらに,複数のモノクローナル抗体を組み合わせることで,現行法と同等の検出感度が得られた.(2)構築した牛乳ELISAキットの検出限界と定量限界はいずれも食品試料中の牛乳タンパク質濃度換算で1.0μg/mL未満であり,原材料表示が必要な数 μg/gレベルの測定には十分な感度であった.(3)添加回収試験では50%以上,150%以下の基準を満たし,現行法の2社のキットと同等の回収率を示した.再現性試験でのCV%は全て10%以下であった.さらに,羊乳および山羊乳以外の食品原材料では交差反応が認められなかった.(4)市販食品における原材料表示と構築した牛乳ELISAキットでの測定結果はすべて一致した.以上のことから,牛乳のβ-ラクトグロブリンに対するモノクローナル抗体のみで構築した牛乳ELISAキットは,特定原材料の牛乳の原材料表示の正しさを検証するための新たな定量検査法として有用であり,また通知法に示された現行法と同等の測定が可能であることが示唆された.
