Abstract
The effect of different plant growth regulators(PGRs) on Taibaimi(Notholirion bulbuliferum) callus induction from bulbules are studied.Fresh underground bulbule of N.bulbuliferum were used as explants for in vitro culture.Surface-sterilized bulbules were incubated on Murashige and Skoog(MS) medium supplemented with different concentrations of 2,4-dichlorophenoxyacetic acid(2,4-D),kinetin(KT),thidiazuron(TDZ) and a-naphthaleneaceticacid(NAA).The results showed that the four kinds of PGRs had very significant effect on the callus induction(2,4-D,TDZ,P0.01;NAA,KT,P0.05).The optimal hormone combination for callus initiation was 0.5 mg·L-1 2,4-D+0.5 mg·L-1 NAA+0.2 mg·L-1 KT+0.1 mg·L-1 TDZ.Some callus could differentiate adventitious buds when they were further subcultured on the same medium.The number of differentiated adventitious buds would increase if KT was replaced with same concentration of ZT in medium.Wild bulbules,callus and adventitious buds were analyzed for total flavonoids content using HPLC,there were no significant difference.They were 13.3%,11.4%,11.9%,respectively of dry weight.
