Research Astragalus Sinicus Seed Vigor with Different Water Content Conserved in the Gene Bank for 20 Years

Abstract
In order to discuss the most suitable water content for long time conservstion,and comprehensive appraise vigor after long-term preservation.This experimental used Astragalus Sinicus seed's with 9.63% water content as material,dried in 35℃ to decrease water content to 8.36%,7.74%,6.92%,6.47% and 5.97%,and packed after seal Tibet to carry under-1℃gene bank condition long-time preservation,after 20 years,took out and determined their germination ability,field growth ability,the conductivity and so on.The result shows: After 20years preservation,the germination rate which with water content 8.36% and 7.74%,6.92% and 6.47% were extremely remarkable compare to before 20years,suggested this type seeds were not suitable long-time preservation;The germination rate which with 5.97% water content were not obviously,suggested this type seeds were suitable long-tiome preservation under low temperature.This result could provide scientific data for the manure germplasm resources long-term safety save and has important realistic significance for preserving germplasm.
