The callus was induced from scutellum of mature embryo of rice(Oryza sativa L.),Then it was subcultured and differentiated properly and studied on the formation of competence and determination.Genotypes played a big role on induction,subculture and differentiation of rice callus.The trend of differentiation competence of rice callus took on down→up→slowly down on subculture.The differentiation competence of the first generation H41 callus kept falling down alone the days passing.When the first generation callus of H41 was transferred into differentiation media after cultured 17d on induction media,the determination could be formed at least inducing 2d on regeneration media.The determination of all the callus could not be formed at the same time because the competence of callus was different.
