Immature embryo scutella of Xindong No.26 were used as explants to study the effect of different combinations of auxins and concentrations on callus induction and differentiation of callus.The result showed that differentiation rate of callus induced by 2,4-D with Dicamba in medium was significantly better than that induced by 2,4-D or Dicamba alone.Differentiation medium containing lower concentration of ZT was propitious to differentiation of root.MS medium containing 2,4-D1.5mg/L and Dicamba 0.5mg/L for callus induction,R medium containing ZT 1mg/L and 2,4-D0.01mg/L for callus differentiation were the best efficiency,the regeneration frequency of plantlets was 90%.On the base of these results,an efficient and stable regeneration system of Xinjiang wheat cultivars was established for biolistics transformation.
