Abstract
AIM To investigate the effect of different dosage forms and concentrations on percutaneous absorption of Draconis Sanguis in vitro,and optimize the dosage regimens of skin with superficial wound.METHODS Modified Franz diffusion cell device was chosen,and needled skin of SD rat which was simulated superficial wound was used as cutaneous barrie.The concentration of dracorhodin in receiving solution was determined by HPLC,and the optimal dosage form and dose were confirmed by calculating the accumulated transdermal rate and the rate constant of percutaneous absorption.RESULTS With the 75% ethanol solution as the solvent,and 5% azone solution as penetration enhancer,the best concentration of Draconis Sanguis was 27.5g·L-1,and the best dose was 82.7mg·cm-2.The optimized formulation was released stably in 8h,and the rate constant of percutaneous absorption J was 0.027mg·cm-2·h-1.CONCLUSION The dosage form of 75%ethanol solution of DraconisSanguisis superior to the other dosage forms,and it can be used as the treatment of superficial wound in clinic.
